There are many similarities between this title and the original Drawn to Life; for instance, three villagers are rescued in levels, and the game's hero, [[DoodlePants]], must clear [[Doodle Scribble]] as they traverse the stage.

The game follows up directly from Frankendoodle. It begins with the clumsy artist once again at sea with two [[Magic Pencil]]s this time, only to drop them into the ocean as he did before. One of the pencils lands on [[Patrick Star]]'s head, while he was practicing his counting, as the other pencil lands a far distance behind him. Patrick remembers that the pencil is a magical one that can bring drawings to life, while trying to remember that there is one thing that he is not allowed to draw. He sketches the forbidden drawing on the ground trying to remember it, which turns out to be [[DoodleBob]], the [[SpongeBob]] caricature and main antagonist from the episode. He springs back to life, stealing Patrick's pencil and running off to downtown [[Bikini Bottom]] in the process. Patrick contemplates what he had done as SpongeBob arrives at the scene, realizing what happened after overhearing him and freaking out. Upon trying to come up with a solution, he finds the second pencil, which he and Patrick can use to draw a hero to fend off DoodleBob's mania. This is where the player is able to draw their hero, which SpongeBob and Patrick name [[DoodlePants]]. Shortly after their creation, Patrick is distracted by a [[Jellyfish]] swarming past and doesn't notice SpongeBob getting captured by DoodleBob's [[Doodle Dude]] army.

After SpongeBob's rescue in the [[Tutorial]], the trio arrive at Bikini Bottom and observe the damages done by the Doodle Dudes. They are approached by [[Squidward]], who informs them that that Bikini Bottom is in complete turmoil ever since DoodleBob's return, and that he stole his clarinet. DoodlePants then must try to bring all of the Bikini Bottom residents back. After four levels, DoodlePants, SpongeBob, Patrick, and Squidward arrive at the [[Doodled Tower]], which was where the Doodle Dudes were being made, and encounter DoodleBob. Squidward demands him to give him back his clarinet, but DoodleBob doesn't understand what he's saying, so SpongeBob gets the idea to have DoodlePants speak to him in Doodle Language instead. After the two Doodles engage in a conversation for some time, Squidward gets tired of waiting and DoodlePob runs off, but not before spawning the first boss in the game, [[Doodled]]. DoodlePants climbs to the top of the building and fights the Doodle robot until it's fully erased. Upon it's defeat, the group realizes DoodleBob drew a rocket and submarine, and use a trail of Doodle tracks to lead them to [[The Jungle]]. The group reaches a drawn [[Pyramid]], which is DoodleBob is revealed to be hiding. DoodlePants goes in to fight DoodleBob, but before he is fully erased (1% left), he runs out of the pyramid and disappears. The group suspects that either DoodleBob used the submarine to go to the [[Deep Sea]], or the rocket to the [https://spongebob.fandom.com/wiki/Moon Moon]. They come to the conclusion that they need to head to Deep Sea first, where they encounter Rock Bottom and the [[Flying Dutchman]]'s Graveyard. 

When they reach the Dutchman's Ship, he approaches the quartet and accuses they are responsible for Doodle Dudes taking over his ship. He is informed of this by [[Doodle Patrick]], Patrick's much more intelligent drawn doppleganger. The group denies this and try to reason with the Dutchman, only for Patrick to mistakenly slip out that he brought DoodleBob back to life. The Dutchman is convinced that it's their fault because of this, and puts the group in charge of getting rid of the Doodle Dudes from his ship. After they're cleared, the Dutchman thanks the group to Doodle Patrick's dismay. Squidward finally reveals that he had tricked the Dutchman, who admits he had some nerve, before he takes over the ship by force. DoodlePants fights Doodle Patrick inside the ship and fully erases him, restoring the ship's control to the Dutchman. He thanks the group and bids them farewell, and they return to Bikini Bottom to prepare to go to [[Space]]. [[Sandy]], who DoodlePants rescued in a prior stage, shows up to thank them for her rescue. SpongeBob asks her to lend them her rocket, only for her to refuse [https://spongebob.fandom.com/wiki/Sandy's_Rocket because of what SpongeBob and Patrick did to it the last time]. She suggests using the Magic Pencil in their possession to create a rocket instead, which the group didn't think of before. Upon drawing one to life, the gang travels to the Moon to catch up with DoodleBob. They encounter [[Bikini Doodle]], a sketched version of Bikini Bottom that DoodleBob drew on the moon, and catch up to DoodleBob a second time. The fight DoodlePants partakes with DoodleBob results in another victory at DoodlePants' end, and the trio celebrates his capture - while Squidward also celebrates getting his clarinet back. While their backs are turned, however, DoodleBob sneaks past them and escapes via the rocket he used before, despite Patrick trying to warn them. DoodlePants chases him once again through space, and lands on an asteroid close to the moon where the final battle, [[Asteroid Battle]], takes place. DoodleBob is gigantic and has an evil, Doodle-coated face and evil red eyes, and sends smaller clones of himself to assist him in fighting DoodlePants off. After Doodlepants defeats him once more, Squidward attempts to erase him for good, only for SpongeBob to realize that DoodleBob created Bikini Doodle because he was very lonely. He gets the idea to erase his mean face and turn him happy, and SpongeBob, Patrick, and Squidward part ways with DoodlePants, who stays behind to keep DoodleBob company. 

After the player beats the 4 main worlds ([[Bikini Bottom]], [[The Jungle]], [[Deep Sea]], and [[Space]]), a fifth post-game world called [[Notebook Land]] is unlocked. In Notebook Land, there are just two stages; the first of which is the last standard level in the game, [[Note World]]. Here, the world can change direction via the Warp Apparatus, which also affects the D-Pad controls, and acts as a gauntlet of all the skills the player learned throughout the game. There is also a boss in this world, [[Last Battle]], where [[Squid Doodle]] - the second drawing brought to life in Frankendoodle - is fought. After the main campaign concludes, the player is free to find and rescue any [[Bikini Bottom Home]] townsfolk they missed, clear any Doodle Goo that they might have overlooked, collect coins to buy color palettes or [[Coloring Book Mode]] and [[Draw SpongeBob]] pages, and replay any level in the game.
